In this study Pleurotusostreus was collected from centre of culture collection CAS in Botany, University of Madras, Chennai, Tamil Nadu. Screening and production of lignin peroxidise, laccase, manganese was carried out. The parameters like pH, temperature, carbon source, nitrogen source and different medium composition were optimized for better production of lignin peroxidase and laccase. The partial purification of lignin peroxidase and laccase was employed by ammonium sulphate precipitation method and dialysed. The partially purified lignin peroxidase and laccase was characterized and the protein profile was analysed in SDS-PAGE and Zymogram. The enzyme production of lignin peroxidase and laccase were more when the medium was amended with glucose and cellobiose as carbon source, urea and ammonium nitrate as nitrogen source. Temperature of 25°C and 30°C, pH of 4.5 produced higher amount of lignin peroxidase and laccase. Additionally studies with medium optimization showed, medium of Olga et al showed higher production of both lignin peroxidase and laccase.
