This book study the effect of some mitochondrial mutations in 16S rRNA, tRNALeu(UUR) and ND1 genes on the incidence of type 2 diabetes mellitus (T2DM) in Iraqi patients. Sequence analysis for the samples revealed twenty seven mutations, among those single-nucleotide polymorphisms, nine variants that were included in the public mtDNA databases (NCBI) were found in this study and were located in 16SrRNA,tRNA Leu(UUR) and ND1 regions. Eighteen novel variants were located in the same genes. M. 3429CT mutations found in T2DM patients more than in normal individuals and showed a significant difference. Many mutations appeared in normal individuals in high percentage more than type 2 diabetes patients and showed a significant difference (m. 3127GA, m. 3139GA, m. 3235GA, m. 3316GA and m. 3480AG). In conclusion in Iraqi T2DM patients there was no correlation between mitochondrial DNA mutations in this study and type 2 diabetes mellitus incidence
