One of the most important developments in scientific research in the recent years has been the possibility of in vitro human 3D cell culture. This technique enable important advances in research, mainly in cancer research, since 3D structures can better mimic the tumor environment when compared to 2D cell culture. In this work, divided in two chapters, we present a review on prostate cancer, 3D cell culture and methodologies for the selection of ligands against tumor cells. Finally, the second chapter, elucidates the application of a very efficient system of 3D cell culture, with the objective of selecting RNA aptamers, specifically against prostate cancer cells. In this way, we developed the 3D Cell-SELEX, aiming to innovate the aptamers selection process, as well as, selecting more specific binding molecules that can be used with efficiency in the diagnose an therapy of prostate cancer.
