Radioimmunoprecipitation assay buffer is a lysis buffer used to lyse cells and tissue, for radio immunoprecipitation assay (RIPA). This buffer is more denaturing than NP-40 or Triton X-100 lysis buffer because it contains the ionic detergents SDS and sodium deoxycholate as active constituents and is particularly useful for nuclear membrane disruption for nuclear extracts. The RIPA buffer gives low background but can denature kinases. A lysis buffer is used for the purpose of lysing cells for use in experiments that analyze the compounds of the cells (e.g. western blot). There are many different kind of lysis buffers that one can apply, depending on what analysis the cell lysate will be used for e.g. red blood cell lysis buffer. Generally, lysis buffers have tris-HCl, EDTA, EGTA, SDS, deoxycholate, and tritonX or NP-40. In some cases it may also contain NaCl(150 mM). In studies like DNA fingerprinting the lysis buffer is used for DNA isolation. Dish soap can be used in a pinch to break down the cell and nuclear membranes, allowing the DNA to be released.
