Formylglycine generation, a unique process necessary for the activation of sulfatases, is a cotranslational event conserved from pro- to eukaryotes and is defined by the modification of the cysteine residue in the CxPxR motif of largely unfolded sulfatase polypeptides to C¿-formylglycine (FGly).
